McFadeyan Reaction
The McFadyean reaction is a staining technique used to detect the capsule of Bacillus anthracis directly in blood or tissue smears.
Developed by Sir John McFadyean in 1903.
Bacillus anthracis has a polypeptide capsule made of poly-D-glutamic acid, which:
-
Is not visible with standard Gram stain
-
Can be selectively stained using polychrome methylene blue (PMB)
The capsule appears as a pink to purplish halo around blue-stained bacilli.
